41 Let us lift up our heart with our hands unto God in the heavens.
42 We have transgressed and have rebelled: thou hast not pardoned.
43 Thou hast covered with anger, and persecuted us: thou hast slain, thou hast not pitied.
44 Thou hast covered thyself with a cloud, that our prayer should not pass through.
45 Thou hast made us as the offscouring and refuse in the midst of the people.

There are many places in the Bible where men are encouraged to pray, to lift up their voice to the God of heaven, to bring their requests before Him, to talk to a God who will hear and answer. Lam 3:41 is one of those places. However this passage also directs us to be sure we are in a position and condition that allows God to answers our prayers. We are to search our hearts and try our ways. (Verse 40) This points to being in a right relationship with God through his Son the Lord Jesus Christ. Unless we are in Christ our prayers cannot be answered. But it speaks as well of our daily walk with the Lord. Israel at that time was in rebellion against God. They had turned to idols, filled the land with innocent blood, were living in a way that was not pleasing to the God they prayed to. As a result God was not answering their prayers, was not moving on their behalf, the nation had fallen to Babylon. What was the problem? It was not God. The problem was hearts pleasing the flesh more than God, hearts walking in the world and not the Spirit. God does want to answer our prayers but we need a daily time of self examination in order to be sure we are in a position that God can do that.
